A blood-based algorithm has cut unnecessary PET scans in an Alzheimer’s trial from more than 70% to 31%. Researchers at USC’s Keck School of Medicine developed the screening model for AHEAD 3-45, the international Phase III trial investigating whether treating people with lecanemab before cognitive symptoms emerge can improve outcomes. Recruitment is difficult because only around 30% of cognitively healthy adults over 65 have enough amyloid accumulation to qualify for trials of this kind. Before plasma screening, more than 70% of candidates receiving an expensive PET scan ultimately proved ineligible. The new algorithm combines the amyloid-beta ratio, p-tau217, age and APOE4 status and reduced that figure to 31%. It was developed using 1,080 AHEAD participants and validated in an independent cohort. It is not a replacement for clinical diagnosis, but its business implications are substantial: blood biomarkers + algorithms could move part of Alzheimer’s detection from expensive specialist infrastructure towards a far more scalable first-line filter. That could lower trial recruitment costs, expand diagnostics and move Alzheimer’s care further upstream towards prevention.
A surprise in ageing biology is warning against overly simple approaches to fighting inflammation. University of Rochester researchers removed the cGAS protein from mice expecting inflammation to decline. Instead, the animals became frailer, experienced more inflammation and had shorter lifespans. cGAS can activate inflammatory signalling through the cGAS-STING pathway when it detects DNA outside the nucleus, but inside the nucleus it also plays a protective role by helping suppress LINE1 genetic elements that can themselves trigger inflammation. The lesson for longevity biotech is important: the same biological target may promote or protect against ageing depending on where and how it acts. This is animal research rather than a human therapy, but it illustrates why future geroscience may require tissue-, cell- and compartment-specific interventions rather than simply switching entire pathways on or off.
Brookdale is providing fresh evidence that demographic demand is beginning to translate into operating metrics for Senior Living. Brookdale Senior Living reported that consolidated weighted-average occupancy increased 50 basis points sequentially in August, while same-community occupancy rose 40 basis points. Month-end occupancy also improved and net move-in activity strengthened. Brookdale operates 532 communities across 41 US states with capacity for approximately 45,000 residents across independent living, assisted living, memory care and continuing care. Executive impact: Senior Living has long been an investment thesis based on future demographics; increasingly, demographics are appearing in occupancy and absorption. For investors, REITs, developers and operators, the coming challenge may shift from finding demand to creating enough capacity, staffing it efficiently and developing products that go beyond the traditional retirement community.
Bogotá is dedicating 1,000 vacancies specifically to workers over 50 today. More than 25 companies and organisations are participating in a September 10 employment fair covering positions from civil engineering, financial planning, accounting and foreign trade to commercial, administrative, technical, logistics and production roles. The city’s economic-development authority explicitly describes the experience, technical knowledge and competencies of 50+ professionals as an organisational asset. Executive impact: senior talent is moving beyond diversity rhetoric into tangible labour-market policy. As careers extend, companies that continue to use chronological age as a filter risk discarding expertise while competitors capture it through second careers, flexible employment, 50+ reskilling, mentoring and multigenerational teams.
Barcelona researchers have also identified a mechanism linking skin ageing to chronic inflammation, potentially changing the future meaning of “anti-ageing”. Scientists at IRB Barcelona found that BMAL1 and YAP, proteins that normally help maintain epidermal function, change their activity with age and begin promoting inflammation-related genes. The study was conducted mainly in mice and supported by human skin data. It also links IL-17 inflammatory signalling to YAP activation; temporarily blocking IL-17 in aged mice reduced YAP-associated activity and expression of the inflammatory genes studied. Translation into safe human therapies remains a future challenge, but the commercial direction is worth watching: skin longevity could evolve from products designed primarily to conceal visible ageing towards precision dermatology targeting mechanisms involved in tissue ageing itself. This creates a growing intersection between beauty, luxury, dermatology, biotech and healthy ageing.
